青枯菌侵染后花生体内保护酶活性及白藜芦醇含量的变化研究

     

摘要

以来自花生重组自交系群体的青枯病抗感家系J109和J112为材料,研究了青枯菌接种后在花生叶片和茎中CAT、POD、SOD活性和可溶性蛋白含量等指标以及白藜芦醇含量的变化情况.结果表明,青枯菌侵染后,在接种叶片中,感病家系J112的CAT和POD活性低于对照或与对照一致,而抗病家系J109分别在第3天起和第1天后CAT和POD活性显著高于对照;在茎中,抗病家系J109的CAT和POD活性显著高于感病家系J112和对照;抗感家系在叶片和茎中的SOD活性与对照相比无明显差异;抗感家系茎中可溶性蛋白含量的变化与CAT和POD活性的变化趋势基本一致.因此,CAT和POD活性可以作为花生青枯菌抗性的一个生理指标.接菌后抗感家系白藜芦醇含量与对照相比均有显著提高,且感病家系J112提高程度更大,说明青枯菌侵染可以促进花生中Res含量的提高,但Res含量的高低与花生对青枯菌的抗性反应无关.%The activities of catalase ( CAT) , peroxidase ( POD ) and superoxide dismutase ( SOD ) , the contents of soluble proteins and resveratrol were investigated in leaves and stems of the J109 and J112 family lines, which derived from the recombinant inbred lines ( RILs) population and were resistant and susceptible to Ralstonia so-lanacearum respectively. The results showed that the CAT and POD activities were similar as the control in leaves of disease-susceptible family J112 after inoculation with the pathogen, while in the disease-resistant family J109,the activities of CAT and POD were significantly higher than that of the control after the 3rd and 1st day respectively. The resistant line exhibited significantly higher activities of CAT and POD in stems than susceptible line and control. No significant difference was observed of the activity of SOD in leaves and stems of the disease-resistant and disease-susceptible family lines. The trends of changes in the soluble protein content were similar as the CAT and POD activities in stems of the two family lines. It was suggested that the activities of CAT and POD could be used to identify the resistance to bacterial wilt in peanut breeding. After inoculation, the content of resveratrol were significantly increased than that of control in the disease-resistant and disease-susceptible family lines,and higher in the susceptible line,which indicated that content of resveratrol in peanut could not be related with the resistance to bacterial wilt, although it could be significantly increased after infected by Ralstonia solanacearum.
