摘要

Surface plasmon resonance is a spectroscopic method of analysis which senses the changes in the refractive index on the surface of the thin metal film. These small refractive index changes can be imaged by using the instrument in an imaging application. In the present thesis, design of the surface plasmon resonance imaging instrument working on the principle of excitation of surface plasmons by prism in a Kretschmann configuration has been described. RecA protein-mediated three strand hybridization and novel DNA microarrays which work on this principle are being studied in our lab. The SPRi instrument would be used in visualization and imaging of this three strand hybridization and studying the properties as well as kinetics of these complexes.
