To understand the expression pattern of the citrus-low temperature gene transcript (CLT-a, and CLT-b) from Poncirus trifoliata during acclimation and deacclimation, RT-PCR and Real Time PCR analysis were performed. The results showed that the transcript level of CLT-a (low temperature induced transcript) starts to accumulate at 10°C and increased as the temperature drops. The stability and production of CLT-a continued as long the plants were kept at low temperature. CLT-b was present with both acclimation and deacclimation at all temperatures, and therefore was considered to be a constitutive transcript. The sensitivity of CLT-a to degradation by temperature was confirmed with an abrupt deacclimation experiment where the plants were transferred, after acclimated, from -1°C to room temperature. In 1 h at room temperature CLT-a levels were reduced drastically.
