Scope and method of study. This study was designed to evaluate whether fixative or stage of estrus cycle would affect biopsy histopathological grade, as well as the representativeness of a single endometrial biopsy sample as compared to samples from three separate sites. Endometrial biopsies were collected in estrus and diestrus from ten mares at the Veterinary Medicine Ranch from the left horn, body and right horn. Each tissue was further divided into 3 parts and fixed in neutral buffered formalin, Bouin's fixative and Modified Davidson's fixative and then histopathologically graded by a pathologist using the Kenney and Doig four category scale. The data were then analyzed using analysis of variance as well as regression techniques.;Findings and conclusions. A single biopsy sample was found to adequately represent the uterus as there was no significant difference in histopathological grade between different sites of collection intra-mare. There was no significant difference between histological grades of samples fixed in the three fixatives; hence it is proposed that Modified Davidson can be successfully substituted as fixative for endometrial samples instead of Bouin's fluid. This would minimize the health and disposal issues associated with both Formalin and Bouin's fluid. A trend of increased histological grades was seen in estrus as compared to diestrus collection of biopsy sample, as well as a significant difference was noted between the two collections. This indicates that the time of collection plays a considerable role in histopathological analysis and reiterates that collection of endometrial biopsy be limited to a fixed stage of the estrus cycle in the mare. This may be of greater importance in cases where paired or multiple biopsies are part of the diagnostic and prognostic plan.
