Cotyledons, stems and leaves of Brassica napus (canola) were inoculated with mycelium of Sclerotinia sclerotiorum isolate Rap-16. Inoculated plants were incubated under humid conditions at different growth stages of the crop; at cotyledon stage, five-leaf stage, and flowering stage. Data indicated that cultivar Zhongyou 821 had better tolerance/resistance to stem rot than cultivar Westar, with the difference significant at cotyledon, five-leaf and flowering stages. Lesion development was quite rapid, 24 to 48 hrs after inoculation. The longer the period of incubation under humid conditions, the more severe symptoms of stem rot were observed. The cotyledon test was the most effective, simple and economical test to identify tolerance/resistance. Rapeseed cultivar Zhongyou 821 exhibits tolerance to stem rot disease. This experiment established the feasibility of using cotyledons and leaves for identification and screening of the S. sclerotiorum-resistant materials in B. napus.
