Quantitative Proteomic Analysis of Amyotrophic Lateral Sclerosis-linked Cellular Stress Response with Tandem Mass Tags

SDS removal spin columns were not needed to remove residual detergent prior to MS analysis; acetone precipitation was sufficient to remove detergents. Use of TMT-6 plex results in 1,800 protein identifications from a HEK-293 total cell lysate, 148 of which are considered to significantlychange between oxidative stress treatment conditions and between expression of WT and mutant (R495X) FUS. The most abundant changes are identified between the WT stressed conditions (0.25 mM SA for 40 min & 0.50 mM SA for 1 hr) as compared to the WT untreated.
