It was developed the immune biosensor for the determination of the anti-insulin level in the blood of patients with diabetes. The next questions were under the detailed consideration: ways of the optimal treatment of the transducer surface to achieve its stable and unified state, main algorithm of analysis fulfillment at the determination of the anti-insulin antibodies in the blood of patients and examination of the developed biosensor in real condition of work. Bare, dodecanthiol and PAA treated transducer surfaces were used in experiments. It was revealed that the PAA is the most suitable for the preliminary stable, cheap and simple functionalization of transducer surface. The main algorithm of biosensor analysis is that when the immobilized insulin interacts with the specific antibodies and the formed immune complex direct and immediately is registered by the SPR based device. To avoid the non-specific interaction between insulin and anti-insulin or other antibodies the samples to be analyzed have to be diluted no less as in 1:100. The results of the immune biosensor analysis correlate with that obtained by the standard ELISA-method. Total time of the immune biosensor analysis may be 5-10 min only.
