The N K edge NEXAFS showed greater intensity of the pi~* peaks at glancing incidence compared to normal incidence. The pi~* peaks in the nitrogen region are likely coupling with the aromatic rings of the bases themselves. The increase in polarization dependence after the backfill indicates that the plane of these DNA base aromatic groups are, on average, oriented more parallel to the surface than the bases on the non-backfilled surface. This can be interpreted as the DNA backbone oriented more perpendicular to the surface when there is the backfilled molecule present, which may improve hybridization efficiency of this surface. This change in orientation may be due to the backfill preventing amine groups present on the bases from interacting with the gold surface.
