Amino acids,the basic unit of protein molecules,are closely related with biological activities.In order to enhance the expression of human-like collagen (HLC),amino acids were added into fermentation broth,and response surface methodology (RSM) to optimize the categories and the concentration of amino acids.The results of PB-design showed that only the dosage of alanine and glutamate had significant effects on the production of HLC.Quadratic polynomial models were established after analyzing the results of CCD-design.The optimal concentration of alanine and glutamate were 3.78gL-1,and 2.52g·L-l respectively.Under the optimal cultivation condition the maximal production of human-like collagen could be up to 0.292g·L-1 which was 11.45% higher than that of the control group 0.262g·L-1 (the highest production of the former study).
