Here we have demonstrated that the MPO-sensitive MR contrast agent, di-5-HT-GdDTPA, is sensitive and specific for active inflammation in the rabbit elastase model of cerebrovascular aneurysm. In the absence of MPO-knockout rabbit models, these experiments validate the use of di-5-HT-GdDTPA as an MPO-specific MR contrast agent in the New Zealand white rabbit. As such, di-5-HT-GdDTPA may be used to study various inflammatory diseases in this animal model. In the current study, MPO, a biomarker of active inflammation in vascular disease, could serve as a biomarker for aneurysm instability and can be visualized using 3T MRI.
