Inhibitory effects of sweetpotato anthocyanins on 60Coγ ray-induced irradiation damage in mouse thymocytes were studied.We first established an jrradiation damage model of mouse thymocytes by using 60Coγ-ray(3650uJ/cm2).The DNA ladders were determined by agar gel electrophoresis.The rate of apoptosis on thymocytes were tested by using flow cytometer (FCM).The scavenging properties of sweetpotato anthocyanins on superoxide anions(O2-),hydroxyl radical (OH·),peroxynitrite (ONOO) and free radicals of lipids were observed by using chemiluminescence and electron spin resonance(ESR)techniques.Results showed that sweetpotato anthocyanins 0.625~2.500 g/l could obviously scavenge O2-(IC50=1.2 × 10-6g/ml),OH·(1050=4.48×10-7 g/ml),ONOO-(IC50=1.0 × 10-6 g/ml) and free radicals of lipids (IC50=4.4×10-5 g/ml)(P<0.05).The rate of apoptosis was significantly decreased by sweetpotato anthocyanins.In conclusion sweetpotato anthocyanins have inhibitory effects as a strong antioxidant on damages of thymocytes caused by irradiation of 60Coγ-ray.
