In vitro permeability studies have been a useful tool to study the mechanisms of drug transport across the oral mucosa, despite of limitations associated with an in vitro measurement. Tissue preparation requires removal of underlying connective tissue. In general, this is carried out using surgical scissors. However, it is a time consuming procedure, which requires a lot of patience in order to successfully isolate the epithelium without making holes in the tissue. Other methods include chemical, thermal and enzymatic treatments in order to split oral epithelium and connective tissue. Typically, in thermal procedures, the intact tissue is incubated in a buffer solution at 60 deg C for 25 min. These conditions seem to substantially alter the permeability properties of the oral tissue(1).
