We report an engineered variant of gall more suitable for clinical translation. Gall was first mutated to resist oxidative inactivation, and further crosslinked via PEGylation to stabilize the bioactive homodimeric conformation. Together, these modifications reduced the effective dose of gall by nearly an order of magnitude. In conjunction with other retention or delivery strategies, we envision that engineered gall dimers will be broadly useful for localized immunomodulation.
