In summary, we have established a highly efficient enzymatic peptide ligation strategy for islet encapsulation and delivery. SrtA reaction was used to permit tunable network crosslinking and immobilization of bioactive molecules (i.e., heparin). Preliminary animal study results showed that euglycemia was achieved in mice receiving gel-encapsulated islets. Future work will focus on investigating the long-term glycemic control in immune-competent animal models using encapsulated islets, as well as studying the effect of VEGF sequestration and release on islet graft vascularization.
