Harnessing Long-Term Release to Produce Robust Long-term Immunity

The primary antibody response to vaccination is greatly enhanced and prolonged with longer-term gel-based vaccine delivery. Moreover, the antibody response following challenge on day 90 after vaccination indicates that gel-based delivery results in both a stronger antibody response (4x enhancement) and the formation of ultra-high-affinity antibodies (125x enhancement). Modeling of pathogen binding utilizing these antibody parameters indicates that increased production of more strongly-binding antibodies following a single hydrogel-based administration allows for complete antigen neutralization during challenge, while typical PBS administration does not yield sufficient neutralization and thus insufficient protection. These advanced materials technologies, therefore, have the potential to provide vastly superior vaccine performance through the precise and sustained delivery of sub-unit vaccines.
