Localized Tacrolimus Delivery for Transplant Rejection Prevention via TyroSphere-loaded Rapidly Soluble Films

Water-soluble pullulan films containing tacrolimus-loaded TyroSpheres were successfully fabricated, sterilized, characterized, and implanted into animal models. In vitro results showed efficient release of tacrolimus from the TyroSpheres in aqueous systems. Target drug concentrations were reached systemically for two weeks in vivo and detectable levels were reached for 6 weeks. Together, these results indicate that the devices provide systemic immunosuppression initially and localized for at least 6 weeks. These thin films can be applied to large tissue beds, allowing for full coverage and rejection prevention in CTAs. Future work will adjust device formulation extend the duration of drug delivery and assess the degree of localized immunosuppression achieved.
