Isolation Identification and Characteristics of Lignocelluloses Degrading Fungus L4

Lignocelluloses on the world are abundant at present, while the usage of them is little. One fungus isolated from wormy lemon, which was demonstrated the capacity of cellulose production from corn bran (as only carbon source), was first reported. The fungus was identified as Neosartorya Fischer by morphological and 18S rDNA sequence, and was named L4. Extracellular enzyme activity of CMC and reducing sugar were assayed. Contents of cellulose, semi-cellulose and lignin in corn bran were examined before and after degradation. In order to obtain the best conditions for cellulose production, the Center-factor composite design (CCD) experiment was used to detect the impact of temperature and initial pH. The software Minitab 15 which is efficient in optimization of various fermentation processes was used to analyze the results of experiments for second-degree polynomial regression fitting. Quadratic polynomial will be got, which will ultimately determine the optimal culture conditions of cellulose production. The current results will be of some help to the screen of cellulose degradation microbe. The related methods of study will improve the efficiency of cellulose production, bring a new class for dealing with pollution and be helpful to the development of cellulosic waste biomass energy in the future.
