Slow-release ointments with nitroglycerin for a safe therapy of anal fissure

Nitroglycerin (NTG) administered topically in analfissure causes side effects due to its good transdermalabsorption. The aim of the study was to develop semisolidpreparation with NTG complexed with on-cyclodextrin(NTG-CD) which will allow for slow release of the activesubstance. The in vitro release studies allowed forconclusion that NTG-CD is unstable in the presence ofwater and anhydrous ointment is the most suitablevehicle. The use of NTG-CD allows for slower release ofNTG from the ointment. Moreover in vitro penetration ofNTG into human skin was slower from the ointmentscontaining NTG-CD complex.
