The construction of organs by tissue engineering using an artificial extracellular matrix have gained increased interest in recent years. In this study, a scaffold prepared by chitosan-alginate polyelectrolyte complex (PEC) was used as an artifical extracellular matrix for tissue regeneration. Epidermal growth factor (EGF) and basic fibroblast growth factor (bFGF) were conjugated with the polysaccharide scaffold via polyion complex. The bFGF and EGF released from the polysaccharide scaffold retained their biological activities as being examined by the in vivo capillary and tissue ingrowth. Obvious cellular infiltration and angiogenesis were shown to occur in the growth factor-conjugated scaffold which was implanted in the dorsal area of rat. The result of thermoanalysis showed that after implanted with the bFGF- and EGF-containing scaffolds, the denature temperatures of newborn collages were close to that of original one. These results suggest that the new scaffold may be useful for the induction of angiogenesis in regenerated wound by tissue engineering.
