Complexes of slightly cationic TAT-liposomesand pCMVβ plasmid encoding β-galactosidasereporter gene (β-gal) have been prepared andadministered into the rat brain in vivo via the carotidartery. Analysis of brain cryosections revealed highassociation of β-gal expression with the endotheliumof the brain blood vessels. In minor extent, neuronalcells were also transfected indicating transfer ofTAT-lipoplexes across the BBB. And therefore,TAT-liposomes can serve as an effective carriersystem for gene transfer into the endothelial cells ofthe BBB and potentially for transfection of otherCNS cells.
