It has been demonstrated that Mesenchymal stem cells (MSCs) are capable of differentiating into neuron and astrocytes. We isolated and purified MSCs from adult bone marrow and induced them to differentiate into neural cells using basic fibroblast growth factor (bFGF) and epidermal growth factor (EGF). Immunocytochemistry shows that after induction for 4 days, a number of the spindle-like cells were neuron-like and expressed nestin. Furthermore, an increased expression of NSE (neuron-specific enolase) and GFAP (glial fiber acid protein) were observed. RT-PCR revealed that nestin, GFAP and NF-M mRNA were expressed in MSCs of the un-induced 4th passages. But the expression of MAP_2 mRNA was not detected. During the induction, expression of nestin, NF-M, GFAP and MAP_2 mRNA increased after 7 days. However, the expression of nestin mRNA decreased thereafter. Among the 3 different induction condition groups, the expression of MAP_2 mRNA were higher in group 1 and 3, but in group 2, expression of GFAP were much higher than group 1. These results indicated that MSCs can be cultivated, proliferated and differentiated into neural stem cells in vitro, which may provide a potential approach for the treatment of neurodegenerative diseases.
