The proposed Enhanced Surface Water Treatment Rule (ESWTR) is considering requirements of up to 6 logs inactivation of Giardia cysts in drinking water treatment. No standard method has yet been endorsed for determination of cyst viability due to complexity and lack of reliability of present methods. Flow cytometric measurements of light scattering properties have been shown to be simpler and more reproducible than current mehods. By pairing monoclonal antibodies and flow cytometry, it is possible to obtain statistically accurate measures of decreasing light scatter over a multiple log range far exceeding the detection limits of methods including the U.S. EPA recommended method of in vitro excystation. The curves generated by light scatter can then be compared to curves generated by other inactivation methods.
