A metabonomic approach based on 1H-NMR spectroscopy, pattern recognition and principle component analysis have been applied for the prediction and evaluation of renal and hepatic toxicity of traditional Chinese medicine. NMR spectra of the endogenetic metabolites from rat urines treated with pyrrolizidine alkaloids (PAs) and cimicidol-3-O-β-D-xyloside (CX), an active component isolated from Cimicifugae rhizoma were measured and statistically analyzed. Toxin-induced changes of endogenetic metabolites could be clearly reflected via the NMR spectra coupled with PCA results. The studies have shown that the renal and hepatic toxicity of the samples could be analyzed simply and rapidly using the metabonomic method.
