The present study aimed to evaluate the potential of nutrients in salmon to ameliorate BDE-47 toxicity in gestationally-exposed mice in terms of gene expression, reflex development and behaviour. Two sets of diets were fed to female mice during gestation and lactation, one set used casein as the protein source and soy-bean oil as the lipid source, while the other used uncontaminated salmon as the main source of protein and fat. Control, low and high (0.5, 6 and 2000 micrograms BDE-47/kg feed, respectively) diets were made for both types of feed. BDE-47 concentrations in pup fat, liver, brain, and stomach content reflected maternal intake. BDE-47 levels were significantly lower in offspring of dams fed the fish-based feed compared to the casein-based feed. Of the 1301genes significantly regulated in pup brains among treatment groups, 1027 responded to increased PBDE in a casein based diet (casein control compared to casein high), whereas 193 genes were significantly affected in brains from pups fed fish-based diets with increasing PBDE levels (fish control compared to fish high). A large fraction of the BDE-47 responsive genes (42%) were also differently regulated in a comparison between the casein and fish control diets. The results of the present study indicate that dietary composition influences the impacts of BDE-47 on tissue accumulation and cerebral gene expression but BDE-47 had only subtle effects on behaviour.
