Polyhydroxyalkanoate (PHAs) is a class of polymers in stored in the cells under the conditions of a nutrient imbalance in the growth of microorganisms. SBR system for the bacteria was established to synthesize PHAs with activated sludge. PHAs was extracted with methylene chloride and chloroform respectively. The amount of the three parallel samples that PHAs extracted with chloroform were 0.125 g, 0.119 g and 0.124 g, relative standard deviation was 3.7%; the amount of the three parallel samples that PHAs extracted with methylene chloride were 0.123 g, 0.117 g and 0.121 g, the relative standard deviation was 3.6%, the relative error of two methods was 2.5%. Experiments show that chloroform can be replaced with methylene chloride for extraction of PHAs.
