To evaluate the effect of toll like receptor (TLR) modulation on the barrier function of respiratory epithelial cells, we have developed an in vitro model of the inflamed bronchial epithelium, using the human bronchial cell line, Calu-3.PCSK (Pam_3CysSK_4), a synthetic TLR2 agonist, was added to the cells under normal and inflammatory conditions. Transepithelial electrical resistance (TEER) and permeability were measured to measure cellular barrier function. To determine the mechanism of action of TLR regulation, expression of tight junctional (TJ) proteins were measured by q-PCR and immunolabeling of individual TJ proteins.Results showed that there is a correlation between the expression of TJ proteins and changes in permeability and resistance of the epithelial cell layer. In addition, TLR modulation is posed to increase the barrier function of Calu-3 cell layers.
