To acquire oligo-peptide as Herpes Simplex Virus subunit vaccine instead of HSV membrane glycoprotein, HSV type-common McAb 1A12 (against gC) which has strong animal protective activity was biotinylated. Using this McAb as selective molecular, a 12 meres phage peptide library was biopanned for 4 rounds. We confirmed the result by ELISa.A conseusus amino acid sequence, -Sg(L)RHII-, has been found by DNA sequencing of 14 clones chosen randomly. The postive phage can react with 1A12 specifically and inhabit interaction partly between 1A12 and HSV membrane antigen, which suggests that the conseusus peptide can mimicry the epitope of gC. This strategy provides the potential for using oligo-peptide as more effective epitope vaccine instead of whole proein, and provides a new approach for developing epitope-specific diagnostic reagents.
