glucose challenge induces oxidative stress in subjects with varying insulin resistance by enhancing CPOA and lowering plasma antioxidant capacity. The extent of oxidative stress was not different between NGT, IGT, and DM subjects. Furthermore, the parameters did not or only weakly correlate with plasma glucose or insulin levels (data not shown), suggesting that the insulin resistant/hyperinsulinemic state per se does not account for the magnitude of changes in the circulating oxidant/antioxidant system. This result questions the hypothesis that glucose-induced systemic oxidative stress promotes the development of macrovascular complications in diabetes mellitus. Possibly, the changes of the oxidative/antioxidative balance in the subendothelium are of more importance in atherogenesis.
